Promising avenue of neurodegenerative disease research may be a dead end

In science, refuting a hypothesis can be as significant as proving one, all the more so in research aimed at elucidating how diseases proceed with a view toward preventing, treating, or curing them. Such a discovery can save scientists from spending precious years of effort exploring a dead end. In a study published in the Proceedings of the National Academy of Sciences, Munich-based researchers refute a widely accepted hypothesis about a causative step in neurodegenerative conditions. These results deal specifically with animal models of human amyotrophic lateral sclerosis (ALS, aka Lou Gehrig's disease) but also raise questions for research on other neurodegenerative diseases, such as Alzheimer's or Huntington's disease.

One of the ways neurodegenerative diseases manifest themselves is in the loss of axons essentially, the transmission lines for electrical signals in individual nerve cells and synapses, the key sites for communication between them. In the past, such damage has been attributed to deficits in the bidirectional transport of organelles, such as the intracellular power plants called mitochondria, along the axons of nerve cells. Now, researchers at the Technische Universitaet Muenchen (TUM) and Ludwig-Maximilians-Universitaet Muenchen (LMU) have put that assumption under the microscope in the most thorough test to date. They used novel imaging techniques to observe changes in both axon morphology and organelle transport with high resolution in both space and time in several different animal models of ALS. Their results show that transport deficits and axon degeneration can develop independently, refuting the hypothesis that one is a direct cause of the other.

The lead researchers in this study were Prof. Thomas Misgeld of the TUM Institute of Neuroscience, a Fellow of the TUM Institute for Advanced Study, and Prof. Martin Kerschensteiner of the LMU Institute of Clinical Neuroimmunology. Together with Misgeld's research group, they observed axonal organelle transport in living tissue in real time and in a way that enabled them to track the movement of individual mitochondria using a novel imaging approach that involves transgenic labeling. They were also able to observe transport of another kind of organelle, endosome-derived vesicles. Several different animal models of ALS were investigated, all of which are based on human mutations associated with the disease.

"The methods both in terms of labeling and in terms of live microscopy that Martin Kerschensteiner and I have developed over the past years to track mitochondria in intact axons now allowed our student, Petar Marinkovi?, to look directly into models of ALS," Misgeld explained. "The result came a bit as a surprise, as we fully expected a problem with transport of these organelles would be present in all the disease models and missing in all the controls. Well, the results taught us differently."

The researchers investigated a number of different questions arising from this surprising result, using multiple animal models of ALS and focusing on motor neurons, the kind of neuron most affected in this disease. By comparison, most previous studies of the putative link between organelle transport and ALS had been limited: Some, for example, focused on a single mutation, made their observations in vitro or in non-motor nerve cells, or did not include a wild-type mutation as a control; others measured surrogates for organelle transport deficits instead of observing transport directly.

Among the conclusions: In ALS models, reduction of organelle transport and initiation of axon degeneration appear to be due to different mechanisms. This suggests that, at least for ALS, axonal organelle transport may be an unsuitable therapeutic target.

In addition, Misgeld said, "We do think these insights have implications for other studies of ALS, or even studies of other neurodegenerative diseases. What our experiments really say is that it is not easy to develop faithful models of neurodegenerative diseases. So it might be worth spending more effort to get better animal models, as this is the only way forward for mechanistic studies, while always checking them against human pathology or human-derived cellular models. In the meantime, it is probably prudent to work with several of the available models in parallel. Moreover, in more general biological terms, our results also speak to the relationship between axonal transport disruptions and degeneration which might not be as tight as we assumed. Here we have a lot more to understand."

Liz Hicks: PAJWLH- Discernment Part 2 and a little fun with ...

So this last month we have talked about relationships for our art journal project. With our last prompt on discernment and understanding how we can better help friendships, with this post I really wanted to talk about friendships...true friendships.?

This is a page that I made to record a special moment that I shared with a friend. Along this same theme, I was asked by my dear friend Courtney Walsh to help her out with the promotion of her new book?

A Sweethaven Summer. Courtney has written two craft books and this is her debut fiction novel. I was fortunate to have met Courtney while she was writing this book so to see it come to fruition is not only exciting for her, it is also exciting for me to see all of her hard work pay off.

When I met Courtney, I was surprised how much we had in common. My family had just moved to Fort Collins in 2009 and she and her family had moved to Fort Collins exactly one year later. Our lives have paralleled to where, Courtney jokingly says that she is living my life, just one year later.? I am a firm believer in things happening for a reason, and since the beginning of our friendship, I felt that pull in my heart knowing that both Courtney and I were brought together for a reason. These reasons have revealed themselves as we have taken turns encouraging each other and cheering each other on in our lives. With our experiences being so similar, it wasn't totally surprising that we had both had our hardships in relationships/friendships as well. These hardships prepared us to see in each other the qualities that we valued in true friendships. I am very fortunate to have a friend like her and I hope you will take a moment to check out her promotion that she is running on her blog.

It is hard to have friends. Sometimes friends take more out of us, than we have to give. Sometimes friends are in a different place in their lives than we are. Friends are not always the blessing on our lives that they are meant to be. I have gotten to a point in my life where I now value the few true soul friends that I have rather than the massive amount of friends that I might have held close in the past. I have realized that good friends are so hard to come by, yet NOT hard to maintain. Good friends don't fault you for time apart, yet they pick right back up where you left off. These friends should be held close and treasured. In my heart of hearts, I want to be the type of friend that loves, gives cherishes my heart and soul friends even if the list of these friends is short. I have learned the hard way that quality is what I need in a friend... not quantity.

So with this page, I wanted to have a little something to remind me of a special moment that I shared with a friend. A time that I was blessed to have this person in my life. This page acts as a reminder in my journal that there are people in my life that make me happy. This page (I will refer to these pages as Little Victories) is just one of the many blessings I want to record in my art journal. There have been to many pages in? art journals that are therapeutic or maybe dark, I really want to be able to look back (or have my children look back) at an art journal that is true to who I am and part of this is recording the good along with the not so good.




So for this prompt, I am going to make it pretty easy :) Here ya go!

Emotion is defined as a mental state that arises spontaneously rather than through conscious effort. It is often accompanied by physiological changes, which means that it affects our body in an uncontrollable way. Our emotions cannot be altered or controlled; we either accept them or release them. Our emotions affect our thinking, our behavior, and our actions. When we have a certain emotion, we will be able to feel it in our body, and it won?t ever go away until we express it or accept it as real. Emotional freedom, now, is being free from feeling like a victim of and controlled by our emotions.

To be emotionally free is to be able to manage our feelings and learn from them.

One way to develop emotional freedom is through emotional freedom technique training. This technique, which is a universal healing aid that rebalances the body?s energy system with respect to unresolved emotional issues, is based on the principles of acupuncture. In other words, it is a form of alternative psychotherapy that uses tapping on acupuncture points while a patient focuses on a specific traumatic memory. This has the effect of clearing the disruption in our energy system. Because the disruption is gone, we are then able to think about that same issue without having the same negative response towards it.

Through Emotional Freedom Technique (EFT), the emotional and the physical reaction associated with the distressing memory or event is reduced by way of neutralizing the connection between the memory or event and the disturbed energy response.

Again, our emotions remain in our body until we find a healthy way to release them. We can achieve this emotional freedom through emotional freedom technique training.

(AP) ? The U.S military announced Sunday that it has recovered the remains of the last American service member who was unaccounted for in Iraq, an Army interpreter seized by gunmen after sneaking off base to visit his Iraqi wife in Baghdad during the height of the insurgency.

The remains of Staff Sgt. Ahmed al-Taie, who was 41 when militiamen seized him on Oct. 23, 2006, were positively identified at the military's mortuary in Dover, Del., the Army said in a statement released Sunday. Army officials said they had no further details about the circumstances surrounding his death or the discovery of his remains.

Al-Taie's brother, Hathal Al-Taie, told The Associated Press the military officer who visited the family's home to inform them about the remains said they are still in Dover, but that he didn't know the circumstances surrounding his brother's death.

"We have no information right now, not even how the body looks like or when they're going to release him," Hathal Al-Taie said by phone from Ann Arbor, Mich., where the family settled after leaving Iraq for the U.S. when his brother was still a teenager.

Their uncle, Entifadh Qanbar, said he was told by the Army major who informed the family in Ann Arbor that the remains were received at Dover on Feb. 22.

"I asked if it was an accident or if he was killed, and he said they didn't know, that they are investigating," Qanbar said by phone from Beirut, where he lives. "He said he had the same questions that I have."

The U.S. Embassy in Baghdad did not respond to a request for comment late Sunday.

Family members say that like many Iraqi exiles, Ahmed al-Taie was eager to help his native land rebuild after the 2003 U.S.-led invasion and ouster of Saddam Hussein.

He met his wife during a trip to Iraq shortly after Saddam fell, while he was still a civilian, and in December 2004 he joined an Army reserve program for native speakers of Arabic and other strategic languages. He was deployed to Iraq in November 2005 and was assigned to a Provincial Reconstruction Team in Baghdad until he was kidnapped the following year.

At the time he was seized, kidnappings for ransom or political motives, mostly of Iraqis but also many foreigners, were common. The February 2006 bombing of a Shiite mosque by Sunni insurgents caused retaliatory bloodshed to spiral, and death squads roamed the streets.

Al-Taie's in-laws say he often met secretly with his wife at her family's home despite warnings that he was in danger of being kidnapped.

It was during one of those visits that al-Taie disappeared. Masked gunmen hiding in an abandoned Saddam-era army building seized him as he went to find his wife at her uncle's house, less than two blocks away in the Karradah.

"A neighbor saw the gunmen and went to my family and informed them. My parents, brothers and sisters all came at once and pleaded with them to let him go," al-Taie's sister-in-law Shaimaa Abdul-Sattar, who witnessed the abduction, told the AP during an interview in 2010 at her Baghdad apartment.

Al-Taie remained calm as he was led into a waiting car and whisked away.

As an American soldier and a Sunni Muslim, al-Taie faced a double risk when he left the protection of his base inside the Green Zone, a well-guarded area that houses the U.S. Embassy, Iraqi government offices and the parliament.

American commanders immediately launched a massive manhunt, locking down Karradah and Sadr City, a Shiite enclave in eastern Baghdad.

Within days, the military arrested four of the kidnappers. But by then, al-Taie had already been handed off to another group and transported to the Shiite heartland of southern Iraq, according to people familiar with the case.

About a week after his abduction, a family member received a ransom demand, the U.S. military previously told the AP.

The relative then met with members of the group behind the kidnapping. They showed him a grainy video on a handheld device of a man they claimed was al-Taie but he demanded solid evidence that al-Taie, who was 41 at the time, was alive and well.

According to the survey, 94 percent of respondents said that they would take one more trip than they already had planned if they could afford it.

The solution, suggests Hotwire, is combining the obligation travel with leisure time. Work sending you to a conference in Orlando? Bring the spouse and kids so they can enjoy some time at the local theme parks.

For each of the first four, I argue that the military means would not respond effectively to the violence, would be far more complicated than advocates acknowledge, and would likely soon pave the way to further escalation upon failure.

I spend?the most time arguing against?the currently fashionable idea of arming the Syrian opposition (about whom, as?Chairman of the Joint Chiefs Martin Dempsey?noted this weekend, little is really known). It is unlikely that arms from the outside would come close to evening the balance of power, and would only invite escalations from Syrian regime forces.? While advocates assume that a better-armed opposition would encourage a wave of defections from the Syrian army, it is just as plausible that growing militarization will harden the polarization in Syrian society and the resolve of Syrian troops.?? Those currently on the fence, disgusted with Assad but afraid of the future, could well be frightened back onto the side of the regime and move even further away from any kind of realistic political solution.

Lynch also argues for sustained economic sanctions and ?enhanced diplomacy? which includes ?constructing such a narrative ? of endless isolation and economic disaster with Asad, or a rapid return to international society with economic revival and political guarantees without him? in order to end the crisis. I can?t endorse Lynch?s take fully, but his arguments against military intervention are insightful.

The link he provided in the above excerpt directs the reader to his previous post which focused fully on why not to arm the opposition. Not only would arming the opposition escalate overall violence, it would also further fragment the already disorganized local militias and its ultimate failure would merely be an invitation for full scale military intervention and even regime change. Also, as we saw in Libya, ?fighting groups will rise in political power, while those who have advocated nonviolence or who advance political strategies will be marginalized.? Lastly, both sides are committing serious crimes, albeit disproportionately, and for god?s sake American taxpayers do not need to be supporting any more human rights abusers.

To me, the broader regional implications of intervening in Syria are enough to invalidate such proposals completely. For those rabid interventionists in Washington, acting against the Assad regime is really just an opportunity to eliminate Iran?s closest strategic ally. Again, war for hegemony in the Middle East would have the deceitful pretext of humanitarian intervention.

HP says 1Q earnings down 44 pct, beats Street

(AP) ? Hewlett-Packard Co., the maker of PCs and printers, on Wednesday said its net income fell, while sales fell 7 percent in the first full quarter under new CEO Meg Whitman.

HP's sales to consumers fared the worst, dipping 23 percent from a year earlier. HP is the world's largest maker of PCs, but it's been performing badly as buyers in the developed world are turning their attention to Apple's iPads and Macs. Meanwhile, it hasn't been able to capture the growing appetite for PCs in emerging markets.

Net income was $1.47 billion, or 73 cents per share, in the three months that ended Jan. 31. A year earlier, it was $2.6 billion, or $1.17 a share. Adjusted for one-time items, the company earned 92 cents per share, above the 87 cents expected by analysts surveyed by FactSet.

Revenue was $30 billion, down from $32.3 billion and slightly below expectations of $30.7 billion.

The revenue drop was even steeper, 8 percent, when taking out the effect of changes in currency exchange rates. It was the fastest revenue decline for the company since the recession hit 2009 results.

CEO Whitman replaced Leo Apotheker in September after a short and stormy reign. Apotheker announced that HP was looking to jettison its PC unit, which has large volume but small profits. That scared off some buyers, who may not have come back when Whitman decided to keep the division.

For the current quarter, the Palo Alto, Calif., company said it expects earnings of 88 cents to 91 cents per share. Analysts were expecting 95 cents per share.

HP shares fell 38 cents, or 1.3 percent, to $28.56 in extended trading, after the release of the results.

Results were buoyed by HP's business of providing information technology services to corporations. The company's second-largest division, after PCs, saw a revenue increase of 1 percent from a year ago.

Sales from the printing division, HP's third-largest, were down 7 percent, and its profitability continued to sag. Sales of printers fell faster than ink and toner sales.

How Did Human Brains Get to Be so Big?

New research points to an ancient energy tradeoff that meant more fuel for brains, and less fuel for muscles.

Recently while visiting the National Museum of Natural History in Washington, D.C., I found myself pondering the noggins of some very, very, old apes.

Along one wall of the Hall of Human Origins ? an exhibit on human evolution that opened in 2010 ? were 76 fossil skulls from 15 species of early humans. Looking at these skulls, one thing was clear: ?millions of years of evolution have given us much bigger brains.

In the 8 to 6 million years since the ancestors of humans and chimps went their separate ways, the human brain more than tripled in size. If the earliest humans had brains the size of oranges, today?s human brains are more akin to cantaloupes.

As for our closest primate relatives, the chimps? Their brains haven?t budged.

With our big brains we compose symphonies, write plays, carve sculptures and do math. But our big brains came at a cost, some scientists say.

In two recent studies, researchers from Duke University suggest the human brain boost may have been powered by a metabolic shift that meant more fuel for brains, and less fuel for muscles.

Our big, hungry brains

Co-author Olivier Fedrigo told me the full story one morning over coffee near his home in Durham, North Carolina. The human brain isn?t just big, he explained. It?s also hungry.

While the brain makes up only 2% of our body mass, it consumes more than 20% of our oxygen supply and blood flow. Compare that to only 7-8% in other primate species, Fedrigo said.

The human brain uses more energy, pound for pound, than any other tissue. Yet our body burns the same number of calories as other primates our size.

In 1995, two researchers in the U.K. published a landmark study arguing that if our overall energy budget didn?t go up, our bodies must have compensated by diverting energy from somewhere else. The theory is called the expensive tissue hypothesis, first proposed by anthropologist Leslie Aiello, then of University College London, and physiologist Peter Wheeler of Liverpool John Moores University in England.

In the years after Aiello and Wheeler published their paper, the expensive tissue hypothesis started to gain support, though scientists had different ideas about which tissues paid the price for bigger brains. ?There?s been a lot of debate about how this tradeoff might have been accomplished,? said Duke evolutionary biologist Greg Wray.

In 2003, an anthropologist at Northwestern University named William Leonard published a study arguing that the price we paid for a bigger brain may have been punier muscles. ?[The thinking was that] even if you?re totally buff, you?re nothing compared to a chimp,? Wray said.

Subsequent studies have raised questions about whether humans are really the scrawny members of the primate clan. Rather than having less muscle than would be expected for an ape of our size, it may be that our muscle is just distributed differently, said Duke anthropologist Christine Wall. Compared with other primates ?humans are scrawny up top but bulky below,? she said.

?But the hypothesis still holds,? Fedrigo added. To build a bigger brain at the expense of muscle, ?our muscles could have evolved to be smaller, or more efficient, or the metabolic cost of walking could have decreased, or it could have been some combination of these things,? he said. ?The possibilities aren?t mutually exclusive.?

In a study published last October, Fedrigo, Wray, Wall and colleagues tested the tradeoff hypothesis and pinpointed changes in two groups of molecules that may have shuttled more energy to our brains, and less to our muscles. ?This is the first time the hypothesis has been tested at the molecular level,? Wray told attendees at the 2011 meeting of the American Association for the Advancement of Science.

The primary source of energy for the brain is glucose, which is pumped into cells where it is needed most with the help of proteins called glucose transporters. Glucose transporters are encoded by a family of about a dozen genes. The researchers zeroed in on two glucose transporter genes, one of which, called SLC2A1, is turned on mostly in brains, and the other, called SLC2A4, is turned on mostly in muscles.

Mutations in SLC2A1 lead to insufficient glucose getting across the blood-brain barrier, which can cause seizures, learning disabilities, or a condition called microcephaly, which turns a normal brain into a tiny one. ?The brain basically starves,? Wray said.

To get a better picture of how these genes evolved after humans and other primates went their separate ways, the researchers compared the human versions of the genes with the same genes in chimps and two more distantly related primates, orangutans and macaques. When they compared the DNA sequences of the genes from each species, they found a number of changes in the human version of each gene but not the other three species.

To find out if those changes may have helped to ferry more glucose to brains, and less to muscles, they measured the amount of mRNA copies of each gene ? a measure of how much protein the gene is likely to make ? in brain, muscle, and liver samples from each species. Compared with chimps, humans make three times more of the glucose transporter found in brains, but only 60% of that found in muscles.

?The tradeoff is exactly what one might predict,? Wray said.

These probably weren?t the only tradeoffs that led to our enlarged brains, the researchers say. In another study published last year, they pinpointed another set of genes that may have funneled more energy to brains, and less to muscles ? this time in the form of a metabolite called creatine. Glucose is the brain?s primary fuel, but creatine provides a backup source of quick-burn energy when glucose runs low.

Creatine is ferried in and out of cells with the help of several genes. When the researchers measured the expression levels of these genes in tissue samples from humans, chimps and macaques, they found that human brains had twice the levels of SLC6A8 and CKB, two genes that regulate how creatine is used by cells. But in contrast to brains, the levels in human muscles were no different from chimps.

Back at the Hall of Human Origins, I turned away from the wall of skulls and made my way to another part of the exhibit, where lifelike busts of eight early humans, outfitted with muscles, flesh and hair, stare out from their cases.

Terror suspect arrested near Capitol in FBI sting

(AP) ? A 29-year-old Moroccan man who believed he was working with al-Qaida was arrested Friday near the U.S. Capitol as he was planning to detonate what he thought was a suicide vest that undercover operatives gave him, officials said.

Amine El Khalifi of Alexandria, Virginia, was taken into custody with a gun that didn't work and inert explosives, according to a counterterrorism official. He arrived near the Capitol in a van with the two undercover operatives, and walked toward the building, according to court papers. He was arrested before he left the parking garage.

El Khalifi made a brief appearance on Friday afternoon in federal court in Alexandria, where a judge set a bail hearing for Wednesday. After his arrest, FBI agents raided a red brick home in Arlington, Virginia. A police car blocked the entrance.

A criminal complaint charges him with knowingly and unlawfully attempting to use a weapon of mass destruction against property that is owned and used by the United States. The charge carries a maximum penalty of life in prison.

El Khalifi, who was under constant surveillance, expressed interest in killing at least 30 people and considered targeting a building in Alexandria and a restaurant, synagogue and a place where military personnel gather in Washington before he settled on the Capitol after canvassing that area a couple of times, the counterterrorism official said. During the investigation, El Khalifi went with undercover operatives in January to a quarry in West Virginia to practice detonating explosives, according to court documents.

He believed he was working with an al-Qaida operative on the plot, according to an affidavit.

El Khalifi came to the U.S. when he was 16 years old and is unemployed and not believed to be associated with al-Qaida. He had been under investigation for about a year and had overstayed his visitor visa, which expired in 1999, making him in the country illegally, according to court documents.

He told acquaintances in January 2011 that he agreed the "war on terrorism" was a "war on Muslims" and that they needed to be ready for war, the affidavit said.

Before settling on a plot to conduct a suicide bombing in the Capitol, El Khalifi considered blowing up an office building in Alexandria, where military officials worked and a restaurant in Washington to target military officials who gathered there. He even purchased supplies that included nails for the operation, according to the affidavit.

Later, when he settled on bombing the Capitol, El Khalifi asked his associates for more explosives that could be detonated by dialing a cellphone number. In January, he unknowingly told authorities he wanted to know if an explosion would be large enough to destroy an entire building.

El Khalifi met with an undercover law enforcement officer, who gave him an automatic weapon that didn't work. El Khalifi carried the firearm around the room, practiced pulling the trigger and looking at himself in the mirror.

A former landlord in Arlington said he believed El Khalifi was suspicious and called police a year and a half ago.

Frank Dynda said when he told El Khalifi to leave, the suspect said he had a right to stay and threatened to beat up Dynda. The former landlord said he thought El Khalifi was making bombs, but police told him to leave the man alone. Dynda had El Khalifi evicted in 2010.

El Khalifi had at least one man staying with him and claimed he was running a luggage business from the apartment, Dynda said, doubting that was true because he never saw any bags.

"I reported to police I think he's making bombs," Dynda said. "I was ready to get my shotgun and run him out of the building, but that would have been a lot of trouble."

Two people briefed on the matter told The Associated Press the FBI has had him under surveillance around the clock for several weeks. They spoke on a condition of anonymity because they were not authorized to speak publicly.

Dar Al-Hijrah Islamic Center imam Johari Abdul-Malik, who along with other Muslim leaders meets regularly with the FBI, said he was contacted by an agency official after El Khalifi's arrest and told the suspect was not someone he needed to worry about.

He said the official told him that Khalifi was "not a regular at your mosque or any mosque in the area."

He said he offered to supply the FBI with surveillance video of the mosque in Falls Church, Virginia, in case it helped with their investigation but was told that was it not necessary.

Police are close to arresting one of El Khalifi's associates on charges unrelated to the terror conspiracy, the counterterrorism official said. The associate was said to also be a Moroccan, living here illegally. Police are investigating others El Khalifi associated with, but not because they believe the associates were part of a terror conspiracy, the official said.
